(2011) uses a spatial light modulator to generate a defined Bessel beam profile that can be swept across a sample and imaged orthogonally. It is a truly elegant solution, and all the information needed to build the device is freely available. WebThe illumination light-sheet usually has a constant beam length independent of the shape of the biological object. Since the thickness and the length of the illumination light-sheet are coupled, a tradeoff between resolution, contrast and field of view has to be accepted.
